چکیده
The purpose of this study was to examine the repairing effect of KLD-12 polypeptide/TGF-β1 nanofiber gel combined with BMSCs in a rabbit intervertebral disc degeneration model. The rabbit models were divided into four groups after 2 weeks: KLD-12 polypeptide/TGF-β/BMSCs gel (A), normal saline (B), KLD-12 polypeptide/BMSCs gel (C), and no treatment (D). X-ray examination revealed that the %DHI (disc height index) of group A increased from 4 weeks to 24 weeks and that these values were significantly higher than those of groups B and C (P<0.05). A similar trend was observed for the intensity of MRI T2-weighted signal. We observed newborn nucleus pulposus tissue in group A, grayish yellow fibrous hyperplasia in group B, white fibrous hyperplasia in group C; and translucent jelly-like tissue in group D. HE staining scores for groups A, B, C, and D were at levels 1-2, 4-5, 3-4, and 0, respectively. The nucleus pulposus cells in groups A and D were deep dyed with toluidine blue. The gray values for aggrecan and collagen II of group A showed slight increases from 4 weeks to 24 weeks, and these values were significantly higher than those of groups B and C (P<0.05). KLD-12 polypeptide/TGF-β1 nanofiber gel combined with BMSCs plays a role in the repair of degenerative intervertebral disc and promotes the regeneration of the nucleus pulposus tissue in the intervertebral disc in rabbits.

Sox9 Gene Transfer Enhanced Regenerative Effect of Bone Marrow Mesenchymal Stem Cells on the Degenerated Intervertebral Disc in a Rabbit Model
OBJECTIVE The effect of Sox9 on the differentiation of bone marrow mesenchymal stem cells (BMSCs) to nucleus pulposus (NP)-like (chondrocyte-like) cells in vitro has been demonstrated. The objective of this study is to investigate the efficacy and feasibility of Sox9-transduced BMSCs to repair the degenerated intervertebral disc in a rabbit model. MATERIALS AND METHODS Fifty skeletally mature...
متن کاملTGF-βl Suppresses Inflammation in Cell Therapy for Intervertebral Disc Degeneration
Recent studies suggest that cell therapy may be an effective way to repair intervertebral disc degeneration. As a strong immune suppressor, TGF-β1 has been shown to inhibit inflammation respond effectively. The objective of this study was to explore the effects of TGF-β1 during bone marrow mesenchymal stem cells-based therapy for disc degeneration. In vitro assays demonstrated that co-culturing...
متن کاملEffect of self-assembling peptide, chondrogenic factors, and bone marrow-derived stromal cells on osteochondral repair.
OBJECTIVE The goal of this study was to test the ability of an injectable self-assembling peptide (KLD) hydrogel with or without chondrogenic factors (CF) and allogeneic bone marrow stromal cells (BMSCs) to stimulate cartilage regeneration in a full-thickness, critically-sized, rabbit cartilage defect model in vivo. We used CF treatments to test the hypotheses that CF would stimulate chondrogen...
متن کاملCo-transfection of adeno-associated virus-mediated human vascular endothelial growth factor165 and transforming growth factor-β1 into annulus fibrosus cells of rabbit degenerative intervertebral discs.
Intervertebral disc degeneration is a common condition that may lead to low back pain and radiculopathy. Understanding the pathophysiology and cellular and molecular events of degenerative disc disease has resulted in the proposal of a gene therapy approach to halt and reverse disc degeneration. We explored the feasibility of reversing intervertebral disc degeneration using human vascular endot...
متن کاملThe role of TGF‐β1/Smad2/3 pathway in platelet‐rich plasma in retarding intervertebral disc degeneration
Recent studies have suggested that platelet-rich plasma (PRP) injections are an effective way to retard intervertebral disc degeneration, but the mechanism of action is unclear. Activated platelets release some growth factors, such as transforming growth factor-β1 (TGF-β1), which positively modulate the extracellular matrix of nucleus pulposus cells. The purpose of this study was to explore the...
